چکیده

Generations of microbiologists have pondered over the issue of bacterial activity and whether most bacteria in the environment are “active,” “inactive,” or “dormant” (Stevenson 1978). This is a question with clear practical implications, in terms of food, sanitary, and clinical microbiology, as well as profound ecological implications. The issue started to be debated in earnest within the aquatic microbial community roughly at the time researchers were developing new methods to estimate the total abundance of aquatic bacteria, since some of the first stains used were thought to respond differently depending on the RNA content of single bacterial cells (see, e.g., Francisco et al. 1973). It was not until bacteria were recognized as being not only very abundant, but also active, that the notion of microbes as important partners in the marine food web gained widespread recognition (Pomeroy 1974). The early studies using radiotracer incorporation showed surprisingly high rates of bacterial

Effect of natural sunlight on bacterial activity and differential sensitivity of natural bacterioplankton groups in northwestern Mediterranean coastal waters.

We studied the effects of natural sunlight on heterotrophic marine bacterioplankton in short-term experiments. We used a single-cell level approach involving flow cytometry combined with physiological probes and microautoradiography to determine sunlight effects on the activity and integrity of the cells. After 4 h of sunlight exposure, most bacterial cells maintained membrane integrity and via...

Physiological and metabolic effects of carbon monoxide oxidation in the model marine bacterioplankton Ruegeria pomeroyi DSS-3.

Ruegeria pomeroyi expresses carbon monoxide (CO) dehydrogenase and oxidizes CO; however, CO has no effect on growth. Nuclear magnetic resonance (NMR) spectra showed that CO has no effect on cellular metabolite profiles. These data support ecosystem models proposing that, even though bacterioplankton CO oxidation is biogeochemically significant, it has an insignificant effect on bacterioplankton...

Influence of an estuarine plume and marine sewage outfall on the dynamics of coastal bacterioplankton communities

Marine bacterioplankton in the coastal region off Aveiro (NW Portugal) develop under the influence of the plume of a mesotrophic estuarine system (Ria de Aveiro) and, more recently, under the influence of the discharges of a marine sewage outfall (S. Jacinto). In an attempt to compare the degree of disturbance introduced by these 2 features to the abundance of sewage bacteria and heterotrophic ...

Near real-time, autonomous detection of marine bacterioplankton on a coastal mooring in Monterey Bay, California, using rRNA-targeted DNA probes.

A sandwich hybridization assay (SHA) was developed to detect 16S rRNAs indicative of phylogenetically distinct groups of marine bacterioplankton in a 96-well plate format as well as low-density arrays printed on a membrane support. The arrays were used in a field-deployable instrument, the Environmental Sample Processor (ESP). The SHA employs a chaotropic buffer for both cell homogenization and...
